Development of an Agarose Gel Electrophoresis Technique for Determining a-Amylase lsoenzymes
نویسندگان
چکیده
Evaluation of a-amylase isoenzymes as a clinical diagnostic aid by previous methodologies has been either too insensitive or too cumbersome for routine clinical laboratory use. With use of an agarose gel electrophoresis system (Worthington Diagnostic Systems, Inc.) serum amylase activity can easily be resolved into at least nine isoenzymes, the resolution being comparable with that of isoelectric focusing. Samples can be analyzed in less than one working day, with use of conventionalreagents.

Electrophoretic transfer as a technique for the detection and identification of plant amylolytic enzymes in polyacrylamide gels.
An electrophoretic transfer technique was developed for the specific identification of isozymes of starch debranching enzyme, alpha-amylase, and beta-amylase. Amylolytic enzymes are separated by native polyacrylamide slab gel electrophoresis and proteins in gels are electrophoretically transferred through starch-containing polyacrylamide gels. Each amylolytic enzyme degrades starch in the trans...

Capillary zone electrophoresis (CZE) and immuno-subtraction electrophoresis (ISE) were evaluated for ability to detect and immunotype monoclonal proteins, compared with agarose gel electrophoresis (AGE) and immunofixation electrophoresis (IFE), respectively. Six hundred seventeen serum samples were analyzed with CZE and AGE to determine sensitivity and specificity in detecting IFE-confirmed mon...

Light-scattering measurements (nephelometry) of serum after filtration through appropriate filters is a rapid method for physically determining triglyceride concentrations and lipoprotein phenotypes. A correlation coefficient of greater than 0.9 was obtained when aliquots of the same specimen were tested for triglycerides by light scattering in an Aminco-Bowman Spectrophotofluorometer and by a ...
